One laughs at the question initially, however after thinking a little more intently about it:
I think the over-arching response would have to be that -- regardless of the fact that it's a very "girlish" bird name -- Tweety is male....but...
Tweety is always portrayed as having long eyelashes, so...is he really?
It's an interesting question, because most cartoons have at least one episode featuring a similar character of the opposite gender that the main cartoon character falls in love with (for that episode, anyway).
This would lend credence to the idea that even with these Warner Bros cartoons they -- maybe -- were trying to make Tweety androgynous. I can't think of another popular cartoon character where that occured (but I could be wrong).
