Personally I think it has to be Eddie Murphy's RAW

Raw and Delirious by Eddie Murphy were stand-up classics, I still wet myself every time I watch them.

The best stand up acts I have seen on DVD in recent years include Peter Kay, Ricky Gervais, Alan Carr, Richard Pryor (well, I saw him on Sky), Dennis Lear, the late great Bill Hicks and even Robin Williams does a great stand-up show. I have never been a fan of people like Chubby Brown or Jim Davidson, I just dont find them funny at all. I tend to like sarcastic humour.

Billy Connelly used to be a great stand up act but now I turn over the channel if I even see his face because during a stand up show - while Ken Bigley from my home town of Liverpool was being held hostage in Iraq - he made jokes including (a) "Don't you wish the terrorists would just get on with it" and (b) made personal jokes about him because his wife was from Thailand. A couple of days later, the terrorists cut of Ken Bigleys head with a boning knife and Connelly refused to apologise. I can understand someone making topical jokes, especially off the bat on a stand up show, but he should have apologised and realised how insensitive it was. I would therefore urge everyone NOT to buy any of his DVD's or even watch him on TV.

Thinking of all the acts over the years, I guess on reflection that Eddie Murphy's Raw is up there as one of if not the best.

Chris Rock is funny!! I have a DVD of his from years ago - has me in stitches every time.

Also like Peter Kay for his very normal everyday life sort of jokes like 'put the big light on' and also Lee Evans purely for the facial expressions!

Eddie's Raw is awesome but I highly reccomend Delirious by eddie as well! (thats the one where he is in a red leather suit!)

But for that kinda humour you really should check out the blueprint that eddie based it on! Thats richard pryor live on the sunset strip!

Eddie izzard is also very funny!

Chris Rock and Lewis Black are favorites of mine. And after reading all the other answers, I'm going to be sure to check out Lee Evans and Peter Kay. I'm not yet familiar with either one.

Peter Kay.

I also bought Paddy McGuinness & Dave Spikey stand up DVDS because of seeing them in Phoenix nights with Peter Kay.
I think I just relate more to northern humour since I'm one myself. Northerner that is, not a humour.
