The antibiotic has to affect something in the bacteria to kill it that does not affect a human. Bacteria use D-alanine to build their cell walls. Humans do not use D-alanine for anything. Pennicillin blocks molecules that bacteria use to carry D-alanine to sites where they are repairing their cell walls. The D-alanine does not get there. The cell wall falls out of repair. The cell wall bursts, and the bacteria dies.

An antimicrobial must be able to the microbe and do something to kill it or inhibit its multiplication. Inhibition of synthesis, or competition with, enzymes is a mechanism. Inhibition of structural peice production or assembly is another.
