Technically, fire is a discovery rather than an invention, or else my vote would be for fire. I'd go with the wheel, too, as a pretty good choice. Agriculture is pretty high on my list of top inventions, because without it, we'd all be pretty hungry (or dead). The printing press is usually listed as being a very significant invention, because it enabled people to share ideas with other people in different places and times.
I saw a survey a while back (last year, maybe?) that asked what the most useful invention was. Choices were the computer, cell phone, car, toothbrush, internet, etc. The most popular answer turned out to be the toothbrush, because people had survived for hundreds of years without all of the modern technology that exists today, but couldn't get by without something as simple as a toothbrush. We may say we can't live without our cellphones, but they're more a quality-of-life issue rather than a true necessity.
Also, modern medicine has saved millions of lives, so I'd vote for "medicine" as a whole, including vaccines.

Telephone.
Without the land line network to connect telephones, we wouldn't have had the network on which the internet was born.
Telephones have evolved into cell phones which are now evolving into internet terminals.

if you consider that we are only here today, enjoying all of the electronic devices that both simplify and comlplicate life so much, ONLY becuase those primitives that came before us were able to survive and reproduce, then I must give it up to those who first fashioned weapons (clubs, spears and knives) to hunt for food and protect themselves from being eaten...without this first step, man would have never dominated earth to rule it...and subsequently, at the intellectual apogee if his existence, destroy it.
