If an agent has a turn over time of 1-2 weeks, there is a possibilty that IF the agent responds back, I will be away. Is it unprofessional to make the agent wait? Or should I just wait until I get back from vacation?

Few agents have a turn around time that short. And even if they do, they do not expect a response within seconds of giving you a reply. Unless your work is time sensitive material, there is zero reason to not send out the query letters now, go on vacation, and if you get a response handle it when you get home.

Depends on how long you're going to be gone. If it's just for a week or two, send out the letters. Most likely it will take them longer to respond, and in case someone does respond while you're gone be sure to leave a message on your answering machine saying when you will be back.

The safe bet is to wait. However, many agents don't make there stated turn around times...

I'd wait until after you return from your vacation just in case. Maybe take some of your time over your vacation to refine and rephrase your query letter so it will be that much better.
