Been years since I read that book but believe George either made a promise to take care of Lennie or just worked with him and decided to become his protector. It made George responsible either way and in the end, George was responsible for stopping Lennie but in this case, it was his love of Lennie that made him kill him rather than have Lennie picked on in prison and killed anyhow.

Lenny's aunt either didn't want him or died, I can't remember, and so George promised to her that he would always take care of Lenny no matter what. I don't exactly remember their relation but I know that they seemed to have a great connection immediately.
