You have to define what is a city. You also have to define what is the first city. You mean historically? Do you mean in rank?

If you were going by culture in the antiquity of man, the first city would be Cairo since the oldest, the longest thriving culture in predominant position was the Egyptian. In western culture it would definitely be Rome since it provides the fulcrum for all European and American culture. All roads lead to Rome.

The Olmec in North America founded cities in North America while most of Europeans clambered around in huts. Recently an archaeologist claimed to have found the ruins of a city on the bottom of a bay in India that goes back thousands of years.

The first city of transport is Rotterdam. The first city of the skies is Atlanta. The first city of diplomacy is New York (United Nations). The first city of money is Zurich. The first city of Peace would be Geneva. The first city of Love is a toss up between Paris and Niagara Falls. You asked such a broad question.

These are all first cities.

Mesopotamia is a region, not a city. Eridu is supposedly the first city in the world, but this will probably never be known for sure.

According to the Bible, it is located in the land of Cain, which is Nod, and probably, this is Eridu, which by then is located at the Persian Gulf. It is in modern-day Iraq.
