I slightly dampen my thumb and forefinger and place the napkin in between them. Lightly rub them back and forth, and the layers should separate. Be careful. Some napkins have 3 layers. Make sure the top layer is the only one you use. It will have no white on the back.

There is a napkin applique glue made specifically for this type of project. Follow the directions on the bottle before altering the napkin itself. Those layers may peel apart, but some thickness is better for adhering to something else. Sample projects are pillar candles, baskets, wood lids on baskets, folders, etc...

I'm guessing you mean a paper napkin. That's what I'll give instructions for. Unfold it and pat it a bit to help flatten it a little. Lick your thumb and first finger and then put one corner of the napkin between your licked fingers. Press together and then quickly pull your fingers apart. The napkin will separate into layers for you. Peel carefully.
