YES!!

I often bring in a He tank for my students to try this. I make them take 3 deep breaths of air between breaths of He so they do not deprive their brains of O2 and pass out on the floor.

I extremely deep water dives He is mixed with O2 to prevent a condition known as "the bends" which can be fatal. This condition is caused by N2 from the air (air is 78% N2 and 21% O2). The pressure of deep dives causes N2 to mix with the blood. The diver must surface slowly and stop at specific depths to get rid of the N2 or they will suffer terrible pain from the bends. By using He mixed with O2 you prevent the bends, but the radio transmissions of these divers is funny to hear. They do not suffer any side-effects from the He, they just sound funny when they try to speak scientific words.

It displaces the oxygen in your lungs. If you do it long enough you will experience a type of light headedness. And if you continue breathing Helium you will asphyxiate and die.
