Spinach does contain iron, which is certainly necessary for strength and muscular development, among other things; but to absorb it you really need to accompany the spinach with food or drink that contains Vitamin C.
The idea of a cartoon character deriving super strength from spinach was actually inspired by a misprint. In a scientific publication of 1870 a decimal point was misplaced, giving everyone the impression that spinach contained ten times more iron than it actually does.
