Simply to many great rock songs to pick the greatest. The two you mentioned are certainly on the list! Love Van Halen but prefer the Hagar years over Roth.

Hotel California. An epic rock song by one of the greatest bands of all time, The Eagles. I could listen to Hotel California a hundred times in a row and still enjoy it. It never gets old. Not to mention, their guitarist is amazing. Hotel California is by far the best American rock song.

I don't think you can limit it to one song, especially since rock and roll has been around for more than 50 years now.
Along the lines of your choice of "Thunder Road", I would have to choose "Born To Run" in the top 25 of the greatest rock songs of all time.
