Yes, the story in Blood+ happens after the events of Blood: the last vampire. No, Saya and Hagi will not end up together in the final episode which is in episode 50 and I would not explain why as I don't like to give spoiler here. The anime has 50 episodes only. Blood+ is an awesome anime which I have grown to like very much. If you have not watch it, I highly suggest that you do.

Yes, Blood+ is a continuation of the movie. Saya developed amnesia and could not remember what happened in Vietnam. The anime focuses on her regaining her memory and fighting off the monsters only she can destroy with her blood.

They are related. Blood+ is set in our time. As you watch Blood+ you will learn what happened in between the movie.
