The local zoning regulations in your town will indicate how many houses per acre are allowed. Some homes are fit on as little as .10 or less; tight and cramped, but still allowable.
Another factor will be if you are on a public water supply (which will allow more homes per acre) as opposed to being in a rural acre that is dependent on a well and septic. The septic system may allow only one home and it will determine the size, i.e. number of beds.
Always good to do your homework in advance. If you're building be sure to prepare your plans in accordance with town/city requirements so that you can avoid problems later.

My mom has a house on .25 acres. I would say 1 decent sized house 2600 sq ft, or 2 or 3 small townhouses with small yards.

Depends upon the size of the houses and the local zoning laws. As a practical matter, that's really just a decent sized single family home lot. If the area is zoned for 7,500 sq ft lots you might get 2 but that would be it for all practical purposes.
