These are what we call card interfaces. PCI is an older version. PCIe x16 is the new slicker model. If you buy a card (ie. graphics card, usb reader, tv card, sound card, etc.) , it usually connects to the computer through an interface. They can either be in the old, slower, but cheaper PCI, or the newer, faster, but more expensive PCIe.
Currently, graphics cards are the main uses of PCIe, just because they need the speed capabilities provided by PCIe. Everything else, in general uses the cheaper PCI.

If that's what the device requires you to have, then it means you need 1 PCIe x16 slot and 1 PCI slot.

For "1PCIe x16", you only need a PCIe x16 slot.

PCIe = PCI Express
PCI = PCI
