That's hard to say, many presidents overcame alot to reach office.
Washington overcame English rule, fought off the English to form a country. ( Can't say that about any other pres, no matter how steller there military record is)
Linclon overcame living in a rural area with out the benifit of the education of those in the cities.
FDR, though he had the political background and up bringing, he had psychical problems that he overcame to become the best president in US history.
JFK, though he was politically connected, he was a Norther and a Catholic. He was also young and a liberal thinker.
Clinton, who was raised primarly by his mother. Wasn't upper class growing up and became president without being well known by the rest of the country.

Franklin Delano Roosevelt
By 1921, it seemed that Franklin Delano Roosevelt a tall, handsome, and privileged young man with influential family and friends had a brilliant future ahead of him. Then suddenly, in August 1921, while vacationing in Canada, tragedy struck. Roosevelt became ill with polio, suffered great pain, and was almost totally paralyzed for a time. Despite great efforts, he never walked on his own again. His mother suggested that he retire to the family's home in Hyde Park, but Roosevelt's wife Eleanor advised him to continue in politics.

Some say Lincoln, who really was raised in log cabins on the then American frontier (the family moved from Kentucky to other western states and ended its migration in Illinois). Lots of candidates claimed that background in order to appeal to voters who admired self-made men, but Lincoln actually lived it.
Some say Nixon. He grew up in a very poor family in California with an alcoholic father. And he was a very shy young man. Those are two very big handicaps to overcome in order to get into politics, let alone rise to the highest office of the land.
Whether or not you agree with his politics or disdain him for Watergate, the guy had to have guts to overcome what he faced.

There are many for different types/degrees of obstacles, which says a lot about the tenacity Americans like to see in our leader...
here's my opinion...
George Washington: obvious, isn't it?
FDR: physical limitation of polio and public perception of limitations
Lincoln: 'Non-privileged' background
Nixon: Introvert, family dysfunction, non-privileged background
Clinton: Family dysfunctions

Abraham Lincoln because he was such an "underdog" especially because of his views on slavery.
