Is it coming from the fan area? Could be that the fan is going in the power supply. If it's a bearing you can actually tell by "banging" on it slightly and it'll change tone or go away for a little bit. Power supplys are pretty easy to change, just make sure everything is turned off and UNPLUGGED!
If it's coming from the front or the area where your hard drive is located then you may want to consider getting your important information off onto another drive or writing it to a DVD. A hard drive crash is difficult to get over.
Take the cover off to identify where the sound is coming from but don't touch anything!

RE:
My computer has started making a loud humming noise coming from the tower. I've sprayed it with air. Any idea
After I sprayed it with air it was okay for about two weeks. Now the noise is back? Will taking the cover off help it?

First thought is that the power supply is going Outside of that, I would check the Coolers on the Video and sound cards. Also the CPU cooler might be an issue. If you open the tower and start the machine, see where the hum is coming from, as well as checking to see what fans are not running. This is usually a good and cheap first start. If no luck there, then I would suggest a Check by a tech

Well these are always hard to answer since I can't look at it, and I can't ask you detailed questions about it. I'll try anyway...could be the hard drive trying to read something and can't and is just constantly spinning. That is bad. It could also be a fan, open up the side and let it run and try to pinpoint the sound, fans go sometimes or just get clogged. It may be the power supply also but my best advice is to try and pinpoint it's source and then go from there. Good Luck.

My computer was making a lound humming noise right before it died. Turns out that it was the power converter. The replacement part costed about $40 at Best Buy.

It sounds like a PSU to me, usually if you open the tower and give it a dust and make sure no wires are catching on the fan it cures the noise, if it persists and if it changes tone or disappears for a second when you hit the top of the PSU.

The fan in the power supply is probably going bad. A new power supply is $20-$30 depending on your type

Remove the cover and tighten all the screws. It is probably your fan but it could be some other moving part in there as well. Also, back up all you data in case it is your hard drive.

The fan may be getting ready to die.
Also, the power pack which has the fan attached is getting ready to go possibly.
We just had this happen & a storm that turned the electric off did it in.
