The way things are looking right now, never. Joe Quesada (Editor in Chief at Marvel)has voiced that he doesn't want Peter Parker with Mary Jane anymore because it limits Spider-Man stories and dates him as in makes him older which isn't a good thing in comics.
However there is the Amazing Spider-Girl comic series which is set in an alternate future where he does have a daughter named May Parker who has taken on the role of the Friendly Neighborhood Web-Head.
As for the movies, not very likely. There is a high possibility they will make more sequels but as for Peter and MJ having kids? Not very likely.

Do you mean in the movies or comics? In the comics he does, May Parker and Benjamin Parker, in the future they both become heroes, Spider-girl and Spider-man II. As for the films is anyones guess.
