You might need to connect something (desktop or laptop, doesn't matter) to the wireless router to complete the initial setup, but once that's done you won't need to leave anything physically connected to the wireless router aside from the cable or DSL modem or router.

I have a wireless network at home, with a cable modem running into a router, which is connected by a cable to one desktop and by wireless signal to the other. As far as i know, the internet service going into the router is all you need to pick up the signal, it doesn't have to be wired into a desktop. You need a modem, a wireless router and something to pick up the wireless signal, whether that's a wireless card in your computer, USB wireless device, Nintendo DS or what have you.

to configure the router to go wireless, connect it to cable first, access the router GUI, enable wireless. after that you can disconnect cable and go wireless. you cant configure a router wirelessly because by factory settings routers have wireless disabled.

all you need is the router to have wireless ability. the only reason you would need to physically connect is to initially set it up to run, even then, most are ready out-of-the-box.
