Yes, there is a difference. A V6 is an engine with 3 cylinders arranged on one side of a "V" with the other 3 cylinder on the other side of the "V". A straight 6 is just that....6 cylinders in a row down the crankshaft. Some think the V6 is smoother running and more powerful, but only putting the engine on a dynometer is going to show it's power.

A V6 has a block that is angled at 90° or 60° and has 3 cylinders on each side. The straight 6 has one long block w/ the cylinders aligned in one straight line. The straight 6 is a very smooth running engine, but I hardly ever see one, I saw one on an old Mustang and recently on a BMW. The smoothest of the V6 engines is the 60° but I think the 90° is more common. I think it just has something to do w/ the ease of manufacturing.

Straight 6 all the cylinders are in line where a V6 they are in 2 rows of 3 shaped in a V.

Another thing is the capacity and engine size. Steright sixes are much longer and bigger than a V6 and depending on bore and stroke, a 3.0 steright 6 will be much bigger than a 3.0 V6.

Additionally, most cars now have the V design, but steright 6 is also a very powerful engine.

The only difference between a V-6 and a straight 6 is the configuration of the engine, the V-6 look just like a V the straight 6 is just straight up and down. Other than that, no other difference.
