I don't remember him singing at all in the movie... did I miss something?

It's an insult, as a Soprano is a young male singer whose voice has yet to change. As such, he's implying that Will Turner is immature and still a boy.

Because Jack thinks that Will is a eunuch (someone who is basically neutered) and some eunuchs do not have there deep man voices, but he a boy voice they are very high and sing like a soprano would.

Jack's implying that Will has less manhood, as if a crucial "part" is missing, so he'd be forced to sing Soprano.
It's just an old expression. ;)

Actually I think Jack's trying to make Will sound gay. I mean, would you date a guy thats a soprano?

i dont think its a singing thing, well sopranos are i think female singers, so its an obvious insult, thats just my opinion.
