A few years ago I took the train (Amtrak) from Los Angeles to Jacksonville, Florida. That trip is on the Amtrak's Sunset Limited train and requires 3 nights on the train. The train left LA at about 9:00PM and we arrived in Jacksonville about 1:00 PM 4 days later.
The Amtrak web site says that due to hurricane Katrina damage they are not running the limited right now. The rout they give requires you to go to DC, then transfer to a train going to Chicago, then transfer to the Southwest Chief which goes to LA.
So you would have to take 3 trains and it would require the better part of five days ( one day to DC, overnight to Chicago, then 3 days to LA).

I like Amtrak expect you can not get from Miami to Los Angeles on one train anymore you will mostly have to take at least 3 trains one to Washington DC then another train to Chicago and then the 3rd train to Los Angeles (or Oakland) and make sure you have plenty of time between connections.. as lots of times the trains run late.. and also suggest you avoid GREYHOUND AT ALL COSTS! they are no longer the good company they used to be they either have a standing room bus which may or may not stop to pick you, you might have to stand for 3 to 4 hours they will leave stranded,..their bus stations are not in the greatest part of town...etc

I've never done it . My best guess would be that it would take 4-5 days. A typical Amtrak routing would take you to DC change trains then to Chicago change trains and then to either LA or San Francisco
