Yes is the proper answer, even though the Czech republic is part of the EU.
If you are from the UK, other photo ID might be enough but the UK is not part of the Shengen Countries, so you will need to use an official document that is approved for border crossing travel, (passport or equivalent.)
If you are not European (USA or other) you will need a passport, and maybe visa too.

Prague is one of the towns worth-visit and to accomplish this you can begin preparing your holiday with hotelbye . Starting with the charming area round the Prague Castle or the Strahov monastery with its remarkable library and finish with …the complete outstanding city, Prague is simply incredible. In Prague you may also visit the little house wherever Franz Kafka worked or the beautiful Prague Castle gardens from where you are able to enjoy a special see of the city with its picturesque red roofs. In that city you will never get bored. If you should go through the spring you will find the spring woods in flower; in the summertime you will see long dusks; fall will provide you with colorful leaves and special light and in winter months you will enjoy the whole city protected with innocent white snow.

Despite the "Yes" answers given, it would appear that some are not aware that European Union Citizens do not need a passport to travel between EU countries. However, for the purpose of having proof of identity, I would take my passport.

You don't need a passport. You need some kind of photographic identification, though. Which is why most people assume you *need* to take a passport.
