I've been there last Summer. We spent 2.5 days there: 0.5 for Great Teton and 2 whole days in Yellowstone NP. I think we saw everything terrific there within 2 days.
Tips: take a look at the map. It marks the important attractions for you. It is very easy to follow the route and the signs are very clear.
What i liked the most:
1. Yellowstone canyon. The colorful canyon is just glorious. Visit there in the afternoon and you are able to take the best picture.
2. Mammoth: hot springs and terrace. You actually can see how a terrace is growing (forming) and dying (disapear).
3. wild animals: keep searching for them, they are everywhere. You will be excited.
I wish I can be there again!

I have been there about 7 or 8 times. When I have gone I have pretty much seen everything in just one day, usually all day. The park is very large and its a long distance between the main attractions. The drive is on a two lane road with just a few places to pass campers and mobile homes. The drive through the park is kinda shaped like a figure eight (8) with the main attractions being on the west side of the park. Then you really need to see The Grand Tetons National Park which is on the South Entrance of Yellowstone. If you have a choice of how you leave the park I do recommend the Northeast exit which goes over Bear Tooth Gap. Very scenic.
Must sees:
-Old Faithful and the Norris Geyser Basin.
-Tower Falls
-Monmouth Hot springs
-The Grand Canyon of the Yellowstone
-Firehole river canyon and falls
-The Grand Tetons
