I think both words could apply and be correct.

I think either word is appropriate for your purposes. They mean the same thing.

First of all, it's grammatically correct to say:
Best wishes for long, happy and healthy life.
Or
Here's hoping your life is long, happy and healthful. (which sounds like you don't quite believe it will be, so I would go with option #1)

both are grammaticaly correct. Healthy just sounds better.

It actually depends on the meaning you are trying to convey, since healthy and healthful have different meanings.
If you are trying to say that you hope their life is one free from disease and infirmity, you want to use healthy.
If you are trying to say that you hope the life they lead is conducive to a state of health and physical well-being, then you want to use healthful.
Healthy basically means free from disease or illness.
Healthful basically means conducive to or tending to create health.
From the context of the rest of the sentence, I believe the word you want is healthy.
