In the US the largest zoo (not counting wild animal parks) by TOTAL land is hands down the Columbus Zoo and Aquarium in Ohio. However not all of their land is developed. As far as total land developed in the US the prize currently goes to San Diego. Yet even in saying that Columbus will pass them by a landslide in the next few years. I believe next year, 2008 after they open a new exhibit on new land they will be the largest by just a few acres, but in 2010-2011 they will blow everyone else out of the water with the addition of just one new area.
Then again I am not sure exactly what you mean by largest... are you talking land or by the size of the collection. Personally I would go by land. You can take a small zoo and cram it full of animals where they do not have much room or a good life, or you could take a large zoo and have a few less animals but in doing so the animals have far more room and better conditions.
As far as the world goes, not sure about that. You might want to check the WAZA webpage for that one.

I am not SURE, but I believe the Toronto zoo is the largest in North America. Toronto houses about 5000 animals to San Diegos 4000. In the USA I believe it is San Diego (again, if you are talking in size meaning animals ... not sure what the answer is if you mean by physical size).
Also, the Bronx zoo makes the claim of being the largest with over 4000 animals, saying that they are the most diverse. HOWEVER, Germanys Berlin Zoo (Zoologischer Garten) has over 1500 species and 14000 animals!!

As North America includes Canada, and they can be found in Alaska as well, I would say that the Polar Bear, able to weigh up to 800 kg might be the largest. Along side the Bison, up to about 800 kg as well. Moose comes in at around 690 kg Grizzlies at about 400 kg. k

That would be Washington, D.C. They have the largest collection of strange and weird animals known, many of whom were bought and brought to the area by lobbyists. They're easily trained to do as their owners tell them, and once they get to D.C., they never want to leave. They even have galleries where people can go and watch their antics, yet still be safe from any direct contact with the attractions.
