Honestly, it probably depends on the "independentness" of the movie. If it's a flat out Hollywood film, I'm sure it's just tobacco or some other substitute, but if it's an independent film, made outside the grasp of Hollywood, there is a chance that marijuana is used.

Really depends on the movie. For a high budget movie, the "legal bud" you might see sold online is often used (like in some of Half Baked). For lower budget movies and television shows, other herbs, like oregano (in Weeds, all but the pilot have cleverly glued oregano was used for dried marijuana).
Marijuana and tobacco are not used in most movies or shows, simply because of the expense and the legal status of marijuana. Not to mention that the actors don't want to be intoxicated while working. Even most scenes with alcohol have water or ice tea instead.

Real pot and tobacco are rarely, if ever used on a movie or TV set. This is due in part to the fact that smoking tobacco indoors is itself banned in most places.
Prop companies sell prop cigarettes, cigars and joints for movie and theater productions. The ones for theater productions often contain incense designed to smell like the real thing.
Oregano and mint are common substitutes, but a mixture designed to be inoffensive is often produced and sold with a name like "herb stage joint" or "Herbal stage cigarette."

They do not use the real stuff. If they did, the production company would have to pay a lot fines, and the producers would be liable for criminal charges.

it's tobacco. If it weren't they could be arrested and the footage used as evidence. But maybe they take a couple "real" rips before filming.
