I prefer the scenic route when taking a trip to a place I've never been to before. The only good part about taking the direct route is to save gas and if I've already been that way there's probably nothing new to see.

Scenic...less of a boring drive. It's better than looking at the same thing. Of course if you're in a rush, direct route.

As already mentioned 10 to 12 hours is a general rule of thumb. That's travelling on the Hume Highway. But you need to take into consideration "city traffic" if you want to travel from (say) Kings Cross to the Bourke Street Mall. You'd then be adding another couple of hours just dealing with peak hour traffic in each of the CBDs. If you leave around 4am from Sydney, you should miss all the peak hour traffic and be on the Hume and heading south without too many hassles. You should then arrive in the outskirts of Melbourne around 3pm in the afternoon, smack in the peak hour traffic. So it will probably take another hour or two to reach the centre of the City.
