Marbella is 595 kms. from Madrid
By car: You take the N-IV road (Carretera de Andalucía) from Madrid south to Málaga (550 kms.), then head west on the coast road (N-340) for another 45 kms. (It's about a six hours drive)
By bus: Go to the ESTACIÓN SUR DE AUTOBUSES. and take a DAIBUS direct Madrid to Marbella. It's about an 8 hour trip. And there's a bus more or less once every hour during most of the day...
By train: Madrid to Malaga you also have several each day. The train may be quicker, but once you reach the Malaga train station, you have to catch a bus from there to Marbella (the train station is a very short walk from the bus station, they are almost back to back).

Marbella has some 100.000 inhabitants. Marbella itself has nothing to offer though, it is not beautiful nor surprising, not very special. But the neighbourhoud ! Drive from Marbella over the hills to the small white towns of Guaro (half an hour)... Alozaina... stop on the way to eat at a Venta... drive further towards Ronda (other half an hour), which everybody loves. If you can , spend the night in Ronda. Obviously: Seville, Granada... If you are into hip go to Tarifa. If you are into quiet and rural, stay in a finca somewhere along the road : horse back riding, hikes in the unesco protected areas... etc etc If you don't mind sitting driving for a few hours: drive from Ronda to Seville, or from Malaga to Granada. I also like Malaga. Humph... too difficult, too much to see :)

Not sure to be honest, but if you go to Marbella, be sure to have a trip to Puerto Banus, which is very close. Well worth a visit and a real eyeopener!
Just looked it up on Google and by bus it takes 8 hours with one stopover.

It's abour 250 - 300 miles and probably 5 hours by train if you take the AVE High Speed Train to Cordoba and then switch to RENFE.
Correction -- it's 396 miles,
