When you "pop someone's cherry," you're breaking a membrane inside the vaginal canal called the hymen. Not all girls have one by the time they become sexually active, as other activities (such as horseback riding or the use of a tampon) can break it without their noticing.

Men don't have any such membrane, and there isn't any physical way to tell whether they have had sex before or not. So, to answer your question, no.
