Behind Enemy Lines was one of the better movies he's made. It wasn't a comedy, and he can be pretty funny, but Behind Enemy Lines was very tense.

I think Owen Wilson was amazing as Lightning McQueen in Disney's Cars. The voice-acting was superb and the role was just perfect for him.

Meet the Fockers
Smallish part, but great character development. Funny movie. All carved from a single piece of wood.

I would have to go with "Behind Enemy Lines" It was the only movie he has done where he is truly acting and not being funny.
