A manuscript is either a hand written or typed, or computer generated original of a book,essay or paper. It can be any length from a few words to many pages. It can be the original work, or it can be a copied work, but usually it is no longer a manuscript when it is published for general circulation to the public.

I am a novelist and manuscript is another word for a novel or a story. I've never heard that being used in an assignment, especially for a public speaking class. Maybe that means he doesn't want traditional essay form. Contact the teacher/professor and ask him to elaborate. He is obliged to answer any and all questions and he is the best person to answer it, as my interpretation may differ from his.
