It is possible to contrive plays that get you a ridiculous score, as in a few thousand points. I have seen a few Web sites with plays like this. Keeping your question within the realm of reality, however, there are these plays:

Highest possible score to hit two triple word scores, with one tile hitting a double letter score (these plays are on the edge of the board) and no other words created: 392 points. The words that can give you this score are: beziques, caziques, mezquits, mezquite, and oxazepam.

Highest possible opening play: 128 points by playing "muzjiks". By the way, the odds that you draw the tiles to make this play are around 55 million to 1.

Note to the others claiming the record for actual play: On the scrabulous.com statistics page given in the next poster's link, the record play is listed as QUEZALES, which scored 431 points. Since this word by itself would score 374 points, another 57 points must have been scored by the formation of other words. Hmm... sounds a little fishy, like maybe it was a set-up?!

The current record high play in NSA sanctioned play is for the word QUIXOTRY which scored over 380 pts.

Edit: Contrary to the answer some places below me, the record setting word is indeed QUIXOTRY. I was however for 365 pts, not more than 380.

Here is a message concerning the record, a copy and paste from a Scrabble newsgroup post.

3. Single Games Scoring Record Shattered -

Michael Cresta scored an amazing 830
points against Wayne Yorra's 490. In the process, Cresta also set
records by scoring 365 points on a single turn (QUIXOTRY) and
averaging 69.2 points per turn and the pair set a new record for
highest combined score with 1,320 points.

National Scrabble Association keeps the official records for game played in Tournamrnt and club play.

Quezales (A great, amaziing play, by any measure) may be the highest-scoring play ever made on Scrabulous, but it is not an NSA record. I stand by my statement of Quixotry as the highest-scoring play eve made in SANCTIONED play.

There is variation in Scrabble that is known as Super Scrabble (I think that's the name!)

In that game, there are two Z's, both worth 10 points each. A word like "SIZZLES" is worth a lot of points, especially when landing on triple word square.

What make this word so enjoyable is also the fact of the two "S"s which allows connecting two more words easier, and getting even more points.

If you don't have any "S"s, then "QUIZZICAL" is also a good word! =)

I was fond of playing scrabble when I was young. I use to visit my neighbor since they own the board game. We would compete and try to outwit each other to win the game. Scrabble helps us to enrich our vocabulary and keeps us entertained at the same time.
It's the best game.

the record for most points actually played was Quixotic, but the word Quizzing (requiring a blank for the second z) is actually worth more points, because when played on the triple-triple location Quixotic was played, the z lands on a double letter score, and is therefore scored 18 times, which is 36 points more than were earned for the x on the same tile.

The words "beziques" or "caziques" would get you the highest possible score if played a cross two triple word scores. 370 points, I believe. One of the letters would already need to be in place before your seven tiles were placed.

By placing "SQUEEZED" on the top left corner of the board,three new words are created:-
"SHACKLES" by keeping S of SQUEEZED ,before "HACKLES" with S on the triple word ;points=51
"ZOOLOGISTS" by keeping Z of SQUEEZED before "OOLOGISTS" ,where one S of OOLOGISTS is a blank ;points=19
"DEJECTIONS" by placing D of SQUEEZED before "EJECTIONS" ,with D on the triple word ;points=60
Points for the word "SQUEEZED" where the second E of squeezed is already available ;S & D of SQUEEZED are placed on Triple words ;points=(27*3*3)+50(for creating a new word using all the 7 letters) ;points=293
Total points=51+19+60+293=423
You get 423 points by placing so.
