Depends on your isometric view. If the Z axis is "up", and X and Y are horizontal (showing at 45º left and right, and if you know the coordinates:
- line x,y,z (first point)
- (next point) @x, y, z

You can also use snap to nodes if you want to join two nodes (the ends of the line you want to draw).

You can also set ucs to front view (xz plane), then view isometric. The line you trace will be in the xz plane... etc

On difficult drawings, I use a layer for nodes only (points) set to PMODE 66 and PSIZE 1, on a bright colour. I set my points in space (point x, y, z), then link with snap in 3D view.

pen or pencil and a straight edge helps
