ive just built a 383 and with the timing on tdc ive had to retard it to get the pining out of it on take off but when it goes in to 3 gear it falls on its face looses all power and claters

Distributor vacuum advance is hooked up to the wrong port on the carburetor.

Unplug the vacuum line and set the timing at idle.
Find the vacuum port on the carburetor that does NOT have vacuum durring idle and plug it in to that one.

ADDED:
There are three types of vacuum:
Manifold.
Ventury.
Throttle.
You need ventury vacuum for your distributor.

Sounds like you are one tooth out on the distributor
1. Remove #1 spark plug.
2. Remove distributor cap
3. Place your finger in the spark plug hole.
4. Bump engine over until compression is felt – observe the direction the rotor is turning
5. Align timing mark @ TDC
6. Check that the plug wires are installed on the cap in the right order.
7. Install the cap

Check your firing order. Could have plug wires swapped.
