My grandfather has a Briggs and Stratton 12 HP Sentennial riding lawn mower. He told me he thinks he needs to grease the gears in the rear end. We took the rear end off and see no plug to do so. Is it possible to do so?

If it is a hydrostatic rear end it is not user serviceable, if it is hydro take it to the shop & be ready to replace the drive unit which is a hydraulic pump motor combination.

You don't say what age the mower is but most of the more recent ones are bolted together and unless the grease has leaked out of a shaft seal they usually don't need grease. They are packed with grease at the factory and turn so slow that there is no heat.

yes u can u just need to follow where the gears lead to and lubricate that area it should take no more then a minute to find the spot
