T minus refers to the time until lift-off; the word minus simply means that it is a counting down, from 10 seconds to 0 seconds, counting by 1.

T- stands for time. Basically they are saying it is time to liftoff minus-or if you take away- this many more seconds.

t represents time equals zero. The engineers set time equal to zero at the occurrence of a specific event, main thrusters turned on, say. Every other event, and every other point in time is compared to the timing of that event. So when they say t minus ten seconds, it's ten seconds before that event.

The "T" refers to time. They count down the time until the start of the mission (at lift off), and then time of the mission begins to count up again until they arrive back on Earth.
