The rear brake line busted on a 2000 Blazer and the valve that shuts the brake fluid shut to keep the rest of the fluid from coming out. How do you reset the valve to open it up again?

Try opening one of the front bleeder screws and leave the back ones closed then depress the brake pedal. Sometimes it takes a good stomp to get the proportioning valve to move back the other way (This is usually due to a slight varnish-type build-up in the valve, sometimes tapping the valve with a hammer will help to unstick it)
GM proportioning valves are spring loaded and should normally re-center themselves unless they stick. Once you get it unstuck it should center itself and then you can proceed with normal bleeding procedures.

They're flow activated.
After replacing the brake line, slowwlly... press the brake pedal down to bleed the air out of the system. (with the brake cylinded bleeder open)

If you push down too fast the check valve activates and blocks the flow.
If you push down realy slow, it shouldn't close off the flow.

After the system has been bled, it should work fine.

ADDED: You may want to borrow a brake line evacuator.

Chilton or Haynes service manual will show how to do that on the proportioning valve. Parts stores carry them.
