She'll have an obvious ovipositor (page down to see photo: http://www.flippersandfins.net/faq.htm#malefemale ) and show vertical stripes through the thickest part of her body: http://www.flippersandfins.net/Images/BettaFAQMGFemale.jpg - note that the stripes won't be visible if your betta is white.

She'll be gravid, which is a fancy way of saying her belly will be quite distended and whitish. If you're keeping her separated from the male by a pane of glass, the male's behavior might become more extreme.
The nest should be large enough, incidentally.
You can let a male and a might-be-ready female together and watch them to make sure neither tries to beat the other one up. If the time is ripe, they'll do their thing before too long, but you might have to separate them if they're really hostile -- the female can be a little offish without concern.
Watch the whole process. It's fabulous.

She will look like she swallowed a marble, full of eggs. She will usually also show breeding bars. If she is ready and you have a male that is ready they will usually spawn in 24 to 48 hours.
