When you reserve an unrelease game at Gamestop, for example, does the reserve add onto the game price, or is deducted from price? Let's say the reserve price is $5.00, and the game price is $40.00.

When you go to buy the game after you have made a reserve, would the game be $35.00 or still be the same price, $40.00?

See it as paying the game in advance. Whatever you put down towards the game will be counted towards the actual price. Gamestop will even let you pay in full if you want.

the reserve price is basically a deposit on the game so it'd go toward the price of the game so the total owed would be $35.00
