If it's classic rock I'd say it would have to be either Queen's somebody to love or Pink Floyd's the Trial. If you want the most influential, though, probably something by Hendrix.

Freebird by Lynyrd Skynyrd
It really is the best. A lot of people deny it because the dislike the whole southern Confederate flag waving aspect of the band (especially disliked by northerners), but if you truly listen to the music you would realize Freebird is by far the greatest classic rock, if not all of music, song of all time.
