One major drawback is the fact that there is nowhere (that we know of) in space to stop and buy more plutonium/uranium to re-fuel your ship. You have to carry all the fuel you need with you. A nuclear fusion reactor would circumvent this problem since you could scoop up hydrogen as you went along to ise for fuel.

The other major drawback, as someone else already pointed out, is that no space launch has a 100% guaranteed rate of success... any sort of malfunction could spread radioactive contamination across populated areas, and that is most definitely a BAD thing.

Two drawbacks are the Columbia and Challenger. Each of these spacecraft exploded over relatively populated areas under their own volition. There is also a Range Safety Officer for each flight that has the responsibility of detonating the shuttle and attached rockets if they are errantly heading towards a populated place.

Any of these causes would lead to a 'dirty bomb' being detonated in the air, possibly the worst way to disseminate contamination/radiation.

Additionally, the heavy shielding required to protect humans from the radiation is prohibitive. This may be offset by the advantages a reactor provides though.
