Just applied plumbers putty to what? Plumbers putty doesn't dry or harden. It's use for things like putting under a new faucet to keep water from leaking back under the faucet and down into the cabinets or under a vanity sink drain to seal the drain. So there is no wait to use it. If your trying to stop a leak around threads, use either Teflon tape or pipe dope.

Plumbers putty is used to seal drain flanges (where the flange meets the sink). It is not made to seal leaking water lines. It is ready to use when the drain flange is tight. (or when the faucet is tightened down to the sink)
