Depends on what age he is.
Also on what your plans are.
Be frank but suited to a child.
They do understand a lot and will always remember the words.
You could say, not now .. maybe some time later.
Or introduce some cousins as a substitute.

I presume that you are not planning on any more children. Maybe getting a friend/relative of the same age to stay over, or a pet. he clearly wants to care for someone/something, a little bit lonely perhaps?

My five year old son is always saying that.
Everyday he asks me for a little brother. (He's even picked out a name for his little brother, lol)
He either wants me to have one, or wants me to adopt one.
I just tell him that I am not ready for another kid.
I try to make him understand how much it hurts to have a new baby(i let him know that he was a c-section baby, and I explained what a c-section is)
I also explain how expensive it is to have a new baby (diapers, bottles, formula, crib, car-seat, etc..)
He is five years old, so he is old enough to understand my words and sentences.
But, if your child is too young to understand things like that, then I don't know what to tell you, hun.

Sign him up for World Vision and let him sponsor an adoptive little brother or sister.

Have him talk to his friends who have brothers or sisters and they will tell him the downside of the little brother/sister deal. My daughter's friends who have siblings long ago convinced her of how great she has it as an only child.

scare the hell out of him or her just tell them, they will no longer be the center of attention and he or she has to learn to share stuff. that will quiet him or her down(example) take a toy from him or her while they are playing with it then they will understand what is the deal after they scream and have a fit you explain thats what you get with a new brother or sister

Do you want him to have one? If so, you could tell him, sure you'll have one coming along soon. If not, just tell him that you are looking for him, and haven't found one yet. He just needs a friend to play around the house that's all.
