For example, I try using it near the hotel resort near Pastoria, (to find the lost key) and it does start beeping - but I really have no idea what I'm supposed to do after that.
Do I continue to walk around until I hit the spot where the item is? Or do I have to press a button/touch the screen?

It is easy to do. Just keep using it. When it starts flashing with more circles it means there is an item close to that quadrant. What you do after is keep using it on that quadrant until you see a black dot on your screen.
