Do it the right way--snail mail a resume, headshot and cover letter. If you haven't heard from them in a few weeks, you may send a follow-up letter or email asking if they have received your materials.

DO NOT CALL THEM! I work in an agency as a receptionist and they get really annoyed. It doesn't make a good impression because they know nothing about you except for what you sound like. It doesn't make you look brave or smart or anything, just annoy and desperate and you don't want that. Mail your headshot and resume with a nice cover letter and it will make a good, professional impression. When it comes to agents you need to show how professional you can be. If you are in a show send them a post card about the show and invite them. The only reason you should call them is to get their mailing address. Do not call them looking for work, they will get very annoyed and hang up on you. I've seen it happen hundreds of times.
