There are several thing you can do, but be aware that if someone wants access they can get it.
1) Buy a padlock for the case, and lock the case so no one can open it.
2) Set the password in your BIOS. Use numbers, letters, and characters. Example: ~123*AbC=
It is very hard to crack.
3) Set you computer where you have to login when it bootup. Many used have it where it goes straight to the Desktop.
4) Set your screen saver to enable the password after a few minutes.
5) Get in the habit of locking your computer when you walk away from it. "Crtl+Alt+Del" select lock.
6) Before you leave for the day go to your NIC's properties page and disable your Network Access.
7) Make sure your computer is set to "Hard Drive Only" in the boot order. the easiest way to access others files is to simply boot from the floppy or CD.

Set the screenSaver on, and check the box that says:
"On resume password protect",
Your screen saver password is the same as your logon password. If you do not use a password to log on, you cannot set a screen saver password.
To lock the computer simple press Control+Alt+Delete and choose lock computer
