I put it at a start price of £150 but now want to revise it, to start price at £50 with a reseve of £150. When i go to do this it says the fees will be about £2.00 is this an additional fee to what the initial fee was? will i have to pay 2 fees due to my initial mistake?

Additional fee, but not to rectify, only because you have to pay to use a reserve

you have up to 24 hours before the item ends to revise it, changing the price will affect the fee by lowering the intial charge, BUT, using a reserve price option will then cost you a fee, the total fee will be shown before you put your item back on the site to sell.
Remember too that after the auction is over, Ebay will take another fee from your final sales price.
