All the answers so far are correct for small dust problems. However, I go a bit further on really dusty computers, I use a leaf blower.
I buy and refurbish used computers, some of which have been stored in closets and garages and barns for some time. So, canned air is usually not effective (and gets expensive if you have a lot of really dusty machines). My leaf blower gets rid of all the dust. Just be "gentle" with it. I muffle the air blast as needed, and I don't aim it directly at the motherboard. Afterward, I make sure all connectors are firmly in pl;ace, because the air blast sometimes dislodges them.
By the way, be very careful wtih vacuum cleaners that have metal nozzels. You can short out your mobo if you aren't careful.

First, you will need to open up the side casing, which is usually held in by two screws that can be loosened by hand. This will open up the inside of the computer so you can get dust out of it. Now to get the dust out, you need to buy compressed air, which is usually available at Office Depot for like 5 bucks. Make sure you put the long nozzle thing on, and blow out all the dust. Make sure to get the fan on the processor too, this is important that this fan isn't clogged with dust. Now just reassemble the side casing and you're done.

Buy a can of compressed air, available at Wal-Mart, K-Mart, Office Depot, Staples, Best Buy, Circuit City, etc., etc., and use it to blow the dust and lint out of the computer. It's best to remove the cover, if you can, AFTER you unplug it from the electrical source. If your tower sits in the floor like mine does, it collects everything from cat hair to dust bunnies. Cleaning it can save you a ton of money!

Go to your local computer store and ask for a can of compressed air.(Used for cleaning pcs) They should know what you're taking about. You can use this to blow most of the dust out of the pc box.

Go to staples, walmart or Radio Shack and get a can of compressed air cost about 4-6 bucks. Open the PC's case (make sure to turn it off and unplug it) and spray away, if its bad you may want to take it outside because sometimes they really have alot of dust. Make sure to spray good around any fan you see inside the case.
That's about it really. You could also use a small electronics vacuum if you want to go that route.

First turn off your PC, use a vacuum cleaner and with small brush get the dust off and the cleaner will do the rest, but dont touch CAPACITOR!!!!
