If you were legally taking the prescription, the employer is barred from asking questions about or testing for the use of legal drugs, because such questions may reveal a disability, which is illegal in the pre-hiring stage. If the prescription is for medical marijuana, they could definitely not hire you - smoking marijuana, for any reason, is a federal offense and they are within their right to not hire someone like that. The first letter linked below states, "An employer also may ask applicants about their current illegal use of drugs but may not ask about their lawful drug use." The second, "An employer also may ask about an applicant's current illegal use of drugs (because an individual who currently uses drugs illegally is not protected under the ADA), but it cannot ask pre-offer whether an applicant currently is taking any prescription drugs, such as tranquilizers, or whether s/he has taken any such drugs in the past."

Edit:
An employer CAN ask after an offer has been extended about what prescription medications you are taking and rescind the offer if the medications would make you unable to perform the essential functions of the job.
