You should think twice, well, make it hundred, before making the decision. Doing that would hurt your sister deeply and don't try using the "thing she doesn't know won't hurt her" excuse because even if she'll never know about it,you do => your relationship with her won't be the same anymore.
Since you said you love her, is that worthy to risk that precious relationship? You can and will have sex with a lot of people, but I doubt that you will find another sister in your life.
Secondly, what the hell is wrong with that brother-in-law of yours? As if cheating on his wife is not bad enough, he has to do it with the most important person of hers too? Sound like he is either a a**hole or he wants revenge on your sister (or maybe you are just too sexy and he can't help it but be an a**hole, but that beside the point).
Seriously though, I feel bad for your sister, having such an a** for a husband. I think you should be on her side and tell him to back off.

Ha, yes go for it, destroy your sister, break her heart into a million pieces and show her she can't trust men or family members.
It's brilliant really, the one thing that can destroy a person.
So go for it, you'll not only not get the guy in the end, but you'll lose your sister. She'll find out, sooner or later, and it will shatter her.
Or be a real man, and realize some things are fantasy only.
